The Spaniard built on his opening round of 67 with a score of 69 to leave him eight-under-par.
Fellow countrymen Pablo Larrazabal and Miguel Angel Jimenez are amongst five players who are just one shot behind.
Overnight leader Matteo Manassero, who fired a course record-equalling 64 on Thursday, hit a 73 to move him down to seven-under.
Ireland's Damien McGrane and Englishman Tommy Fleetwood are both also one shot back from De la Riva.
