With a victory target of 430, South Africa began the day on 77-4. 35 overs later that total had become 126-4.
Du Plessis, who has had two successful lbw appeals against him overturned via DRS review, is on 49 not out.
Meanwhile, De Villiers is unbeaten on 31.
