The result has seen the Strikers secure themselves a home semi-final this weekend, while the Renegades are out of the competition.
Adelaide made 166-5 from their 20 overs before Melbourne fell short with a score of 144-7.
Travis Head (71) and debutant Kelvin Smith (41 not out) both made big contributions with the bat for the away side, who also had Craig Simmons (15) and Johan Botha (32) do some damage at the crease.
Nathan Rimmington ended with bowling figures of 2-40 for Melbourne, with James Pattinson and Fawad Ahmed each finishing with one wicket apiece.
The Renegades put up a valiant fight out in the middle, but they struggled to keep up with the required run rate.
Matthew Wade (22) and Ben Stokes (33) dug in towards the top of the batting order, and they were supported later on by Shakib Al Hasan (22) and Tom Beaton (23 not out).
However, the Adelaide bowlers took wickets as regular intervals as Ben Laughlin and Adam Zampa both claimed two victims each. Gary Putland, Kane Richardson and Botha also got in on the act with the ball.
