Villegas heads into day two with a one-stroke lead over William McGirt and Webb Simpson after sinking five birdies and an eagle in an impressive opening day in Greensboro.
Britiains Paul Casey and Martin Laird also enjoyed stellar opening rounds after both finished Thursday on five-under-par.
Englishman Casey managed a bogey-free round, recording five birdies en route to a 65, while four birdies and one eagle will also see Scotland's Laird trailing by two shots at the start of play on Friday.
