Wawrinka enters the tournament looking to win his third Grand Slam title, but at times during the opening two sets he struggled to find his range against the Spaniard.
The Swiss number two found himself a break down in both sets, but turned things around to establish a commanding lead over the world number 58.
Ramos-Vinolas refused to fade in the third set though, and eventually broke his opponent as momentum appeared to switch in the 27-year-old's favour.
Wawrinka showed his class to break the Spaniard back immediately though and enforce a tie-break, which the favourite won 8-6.
