After the withdrawal of captain Wayne Rooney, his Manchester United teammate Jesse Lingard will come into the side to form an attacking-midfield three alongside Adam Lallana and Raheem Sterling.
Jamie Vardy is also given an opportunity to end his scoring drought for both club and country after being selected in place of Daniel Sturridge.
The other change sees Nathaniel Clyne come in for Kyle Walker at right-back as Southgate opts against mass tinkering after his team's 3-0 win over Scotland on Friday night.
As for Spain, Chelsea defender Cesar Azpilicueta and Manchester United and Manchester City playmakers Juan Mata and David Silva all feature in Julen Lopetegui's team.
There is also a rare appearance in goal for former Liverpool stopper Pepe Reina.
