Alessandro Florenzi then completed the scoring just before half time with a brilliant counter-attacking goal, which saw Roma find the net on the break from an Inter corner.
Inter did little to threaten a comeback in the second half, although Andrea Ranocchia had a header harshly disallowed for an apparent infringement involving goalkeeper Morgan De Sanctis.
Roma were forced to play the final 11 minutes with 10 men after Federico Balzaretti picked up a second yellow card for a needless foul on Ricardo Alvarez.
Despite their numerical disadvantage, Roma remained untroubled and extended their lead at the top of the table to five points.
Roma are now unbeaten in eight matches against Inter, a run that stretches back to May 2011.
