The 19-year-old, who has made 33 appearances for his Argentine club this season, is widely regarded as one of the brightest talents in world football.
"He's a proven player in Argentina despite his youth, he was captain of his team and is overflowing with talent," Valencia general manager Rufete said in a statement.
"We believe Rodrigo has everything [required] to succeed in Valencia and European football."
It is understood that Valencia have parted with £5.3m for his services.
