The hosts appeared on course to end their four-match losing streak and led for 83 minutes after Luc Castaignos put them ahead early on.
The reported Premier League target ran on to Hakim Ziyech's pass to notch his 10th of the season with just five minutes on the clock.
Twente could not find a second goal despite their 58% possession and 14 attempts on goal and were made to pay for misses by Castaignos and Jesus Manuel Corona late on.
Adnane Tighadouini scored a double to secure last week's victory over Cambuur and turned provider to supply substitute Falkenburg with the chance to fire home the leveller.
